Summary:

The Senior Principal Scientist / Principal Scientist is responsible for developing novel chemical processes for ADC drug-linkers for each development stage, from pre-clinical to commercial manufacturing, of ADC therapeutic agents. The ideal candidate will be expected to leverage their expertise in process chemistry to design chemical processes for a wide range of chemical transformations, identify reaction parameters which impact scale up and purity profiles of complex reaction products, and to explore all viable techniques for purification of target molecules. In collaboration with Seagen colleagues across the research and development, this position will also contribute to solving cross-functional challenges in the development of novel ADC drug candidates. The ideal candidate would contribute to the expansion of external development and manufacturing partnerships to ensure that state of the art technologies are employed in the development and manufacturing of novel ADC drug linkers. The candidate would also be expected to leverage their industrial and academic professional network to enable the continuing improvement of the capabilities and expertise within the Process Chemistry department, and to the general advancement of ADCs as a growing class of drug therapy.

Responsibilities:

  • Demonstrate exemplary scientific leadership through the evaluation and design of synthetic routes to complex molecules which would serve as the foundation of efficient processes for the preparation of multi-gram to kilogram scales of ADC drug-linkers.
  • Apply the latest technologies in both organic and analytical chemistry to develop and interrogate chemical processes while capturing critical data for the determination of reaction parameters that contribute to process performance and scale up.
  • Using established principles of chemical process development, design efficient processes with facile operational steps for execution in conventional pharmaceutical manufacturing facilities.
  • Serve as the technical lead of Process Chemistry project teams in the development and transfer of complex chemotypes to third party contract manufacturers, and in collaboration with Process Engineering Leads, provide continuing technical oversight to ensure successful execution of processes at scale.
  • Contribute to the career advancement of process chemistry staff members by providing technical guidance and mentorship.
  • Build and leverage an external academic/consulting network and identify opportunities for their contributions to process chemistry development challenges.
  • Contribute to organizational knowledge capture. Serve as author and reviewer of departmental technical documents to ensure appropriate capture of process knowledge in support of CMC team development and regulatory strategy.
  • Identify new principles and concepts from other fields to promote progress in synthesis and process chemistry.
  • Contribute to Seagen’s position as an industry leader in the development of ADC technologies through external publications and presentations.

Qualifications:

  • Minimum of 10-15 years relevant industrial experience (PhD) or 15-20 years (BS/MS) in developing synthetic routes and optimizing chemical processes for active pharmaceutical ingredients.
  • Possess a high level of expertise in organic chemistry, reaction mechanisms, and knowledgeable of current advances in the field.
  • In-depth knowledge of ICH/FDA regulations and current industry GMP guidelines for pharmaceutical manufacturing.
  • Proven track record in resolving cross-functional CMC issues in small molecule development.
  • Experienced in the transfer chemical processes to external CMO and CROs and oversight of external GMP manufacturing.
  • Excellent written and oral communication skills; enthusiasm, integrity, self-motivation, and ability to interact across research and development organizations.
  • Prior project leadership experience in oncology / high potency APIs is preferable.
